The city of Bayreuth is located in the northern part of Bavaria in a picturesque region with the highest density of beer breweries worldwide. Within 1 and 2 ½ driving hours you can reach the medieval city of Nuremberg and the Bavarian capital Munich, respectively. The University of Bayreuth (www.uni-bayreuth.de) is a public university focusing on inter¬national collaboration and interdisciplinary research. The university has an outstanding repu¬tation in a broad range of disciplines and currently maintains a network of more than 450 international partnerships with research institutes and universities around the world. The Institute of Cell Biology uses the baker’s (and brewer’s) yeast Saccharomyces cerevisiae as model organisms to elucidate mech¬anisms of mitochondrial morphology, dynamics, inheritance, and degradation (www.zellbiologie.uni-bayreuth.de; Westermann B., Nat Rev Mol Cell Biol 2010). Our working is integrated into the Institute of Cell Biology, aims to dissect conserved cellular mechanisms of programmed cell death in yeast models for human neurodegenerative disorders. Our work is focused on the role of mitochondria and protein degradation pathways in modulating cytotoxicity of human neurotoxic proteins in yeast, including amyotrophic lateral sclerosis-associated TDP-43 and Alzheimer disease-associated aberrant ubiquitin (UBB+1).
In our lab we currently established novel genome-wide screening methods, mainly based on protein fragment complementation, to identify yeast interaction partners of human neurotoxic proteins like TDP-43 in vivo. By elucidating unknown interactors of TDP-43 we expect to get a better understanding of cellular factors and pathways involved in TDP-43 localization, toxicity, and clearance in yeast and further transfer those findings to the human disease model. Your task will be to employ and optimize different approaches to find novel TDP-43 interaction partners and characterize their impact on TDP-43 in a cellular context.
You will apply advanced knowledge and practice in:
Molecular cloning techniques
Immunoprecipitation and western blotting
handling of yeast cells
fluorescence microscopy
addressing cell biological problems with adequate molecular proceedings
