This is a Trialect partnered Traineeship program that is exclusively available to the registered members of Trialect. This program includes research on patients as well as in the lab;

We are experts in the investigation of red blood cell volume regulation, detection of density, deformability and metabolic state of cells. We are interested in detection of cytoskeletal disorganization and defects associated with facilitated proteolysis in red blood cells. We have experience in working with blood samples of health humans and patients with hereditary rare anemias (sickle cell disease, spherocytosis, xerocytosis, cryohydrocytosis, PK and G6PD deficiencies and further idiopathic hereditary disorders associated with hemolysis and increased red cell turnover or inefficient erythropoiesis. Measuring intraerythrocytic free Ca2+ levels, membrane permeability to Ca2+ and function of Ca2+ pumps as well as following on Ca2+-dependent processes in red cells is our specialty.

We are the leading house for the RELEVANCE project on Regulation of red cell life-span, erythropoiesis, survival, senescence and clearance (http://relevance.arivis.com/) and a partner within the CoMMiTMenT project network on Combined Molecular Microscopy for Therapy and Personalized Medication in Rare Anaemias Treatments 

We are routinely applying the following experimental techniques:

  • Na+, K+ intracellular and extracellular concentrations (flame photometry, gravimetry, ion-selective electrodes)
  • K+ loss, glucose uptake, and lactate production rates in RBC suspension/whole blood (electrodes, enzymatic micro-assays)
  • Cell density, volume, hydration state: Percoll density gradient, microfluidic test, (intracellular Na+K), morphometry
  • O2 dissociation curve: HEMOX analyser
  • Ca2+ movements and intracellular content: fluorescent dye fluo-4 AM (micro-fluorescence imaging – microscopy and flow cytometry)
  • Filtration assay (deformability, membrane stability)
  • Cytoskeletal integrity (microscopic test and SDS page)
  • GSH/GSSG – Ellmann’s reagent, glutathione reductase
  • ATP: bioluminescent test
  • NO2-/NO3-: chemiluminiscence test (CLD-88)
  • We may detect activity of single ion transport systems (Na,K-ATPase, Na,K,2Cl- and K-Cl- cotransporter, Na/H exchanger, Gardos channels, NMDA receptors) using radiolabeled tracer kinetics and radiolabeled antagonist binding assays
  • To come soon: microfluidic red cell adherence technique

Me and my mentor (Prof Dr. Anna Yu. Bogdanova) have collaborated already and have applied for a funded research project on the area of specialization. It was a great professional opportunity to be a part of the Clinical Translational Research-Anemia and Red Blood Cells Traineeship. Prof Dr. Anna Yu. Bogdanova and her research team were very supportive and great teachers. In my laboratory, we have established the techniques learned during the program. I have learned the most appropriate techniques needed to achieve the objectives of the ongoing research. We are in the path of developing novel protocols for the early prenatal identification of genetic diseases using the techniques learned from the Trialect Traineeship. The program was well designed and executed appropriately. I am okay for the future fellows to contact me.
