Advanced Optical Microscopy in Biomedical Research

In the first week, the candidates would have to dive into literature on the imaging techniques and the specific topic. If they arrive before 7th of June they can follow a dedicated course on the imaging techniques that will be given in week June 7 to 11. In the first and second week, we will teach them to handle the microscope and do experiments on their own. Since obtaining ethical protocols for animal experiments is not feasible within the short period for the project, we will refrain from in vivo animal projects. We have several ongoing projects that could be interesting for the candidates. Examples are mitochondrial imaging, imaging of the nuclear lamina in laminopathies using super-resolution microscopy, uptake of extracellular vesicles by Endothelial cells, imaging of cleared tissue, imaging of human placental or epilepsy tissues, etc. The candidates embark on those specific projects in week 3, continuing till week 6 or 11, after which they will have to write a scientific report, summarizing the data. If enough valuable and valid data can be gathered, these could be included in publications.

The methods used will be Nuance Spectral microscopy, Stimulated Emission Depletion (STED) super-resolution and Confocal microscopy, and Two-photon Laser Scanning (lifetime) Microscopy.

3D image restoration and image processing

The equipment is installed in the CORE lab of Maastricht University and is part of the EuroBioImaging site “Maastricht Excellence in Imaging”.

Although I could follow the students' projects and learn in practice about the techniques, I think it would be best if there were exclusive prepared samples for the traineeship. I stayed for 6 weeks at University of Maastricht for the Advanced Optical Microscopy in Biomedical Research. The professors/mentor hosts that give the traineeship have a lot of experience and knowledge about microscopy, they were friendly, helpful and open to feedback. I was able to follow the students' projects, learn the routine of the research group and experience real life problems and solutions that one could encounter in this field of research. I enjoyed my time at the University of Maastricht, all the staff were super helpful, and I was able to meet very talented researchers and students. I already had some experience with microscopy, but after this traineeship I’ll be much more confident to decide which technique to use in my nexts projects, and I think that is the most valuable lesson that I learned from this course. Overall I think it was a very positive experience that met my expectations. I was very well received by the mentor host research group. I felt part of their team for these 6 weeks. I also had the opportunity to use high-end equipment that I don’t have access to in my local region. The Netherlands and especially the city of Maastricht were very pleasant and definitely will be missed. Although I could follow the students' projects and learn in practice about the techniques, I think it would be best if there were exclusive prepared samples for the traineeship.
